Foundayo (orforglipron) is a once-daily oral GLP-1 capsule for weight loss and long-term weight management in adults with obesity or overweight and at least one obesity-related condition. It works by reducing appetite and food intake.
Indications: Who is it for?
For adults with obesity or overweight and at least one obesity-related comorbid condition, alongside a healthy diet and physical activity.
How to Use
Take once daily at any time, with or without food. Swallow the capsule whole; do not open, break, crush, or chew it.
Dosage & Administration
The starting dose is 1 mg once daily. After at least 30 days, it may be increased to 3 mg, then to 6 mg, 12 mg, 24 mg, or 36 mg. The maximum dose is 36 mg once daily. Do not take more than one capsule per day.
Warnings & Precautions
Use with caution in patients with a history of pancreatitis or severe gastrointestinal disease. Foundayo is not recommended in severe hepatic impairment. Its safety and effectiveness in children under 18 have not been established. Effective contraception is recommended during treatment.
Possible Side Effects
The most frequently reported side effects are gastrointestinal disorders, mainly nausea, vomiting, and diarrhea. These were more common during dose escalation and decreased over time.
Drug Interactions
Strong CYP3A4 inhibitors may require an alternative medicine or a reduced Foundayo dose. The maximum dose with a strong CYP3A4 inhibitor is 12 mg. Moderate or strong CYP3A4 inducers may reduce its effectiveness.
Contraindications
Do not use Foundayo if you have a personal or family history of medullary thyroid carcinoma (MTC), MEN 2, or hypersensitivity to the active substance or any of its excipients. It should not be used for weight reduction during pregnancy or during breastfeeding.
Storage Instructions
Store at or below 30°C in the original package to protect from light.
